About GANX

Gain Therapeutics, Inc. is a redefining drug discovery company, which identifies and optimizes allosteric binding sites that never targeted before. Its See-Tx target identification platform uses the published 3D structure of enzymes and a proprietary computational technology to discover new allosteric binding sites and predict their druggability. The firm is also unlocking new treatment options to treat disorders characterized by protein misfolding. The company was founded by Khalid Islam in 2017 and is headquartered in Bethesda, MD.

$GANX Khalid stepped out from behind the curtain yesterday. I said he would only do this if and when he knew he has it. This is a man who speaks with intent and purpose. Nobody has more integrity and his reputation is everything to him, next to helping the people who benefit from the drugs he has been involved with. This is a man whose pride possessions are the letters he has received over the years from both patients and their family members thanking him. Nobody knows rexacerat better and his post yesterday likely goes down as a timely tell. $XBI

$GANX If you are a Parkinson's patient, or your loved one is, realize that there is currently no other drug out there in development, that does what rexacerat does for them, nothing. Look high and low, it doesn't exist. The stock price while disappointing and absurd, is only further solidifying my opinion that the company likely has a new name on the door in the near future. Especially with thisnweeks conference, others and Jefferies coming up by year end. As I said before, shareholders are likely to play a key role with their votes determining if an offer is approved. The offer is likely to come with CVR's which we will have to factor into the equation. Some perspective, Vanqua is said to have a valuation of nearly 10X ours. They have a single drug that hails in comparison to rexacerat, let alone the fact we have another even possibly better drug right behind it that could be in the clinic in less than 2 years itself with rexacerat possibly being approved with a phase 2 being pivotal. In addition, they also have a handful of other compounds already likely laying in wait maling it a complete platform play with a large pipeline. This is literally the perfect opportunity for almost any major pharma due to the clear price disparity and pipeline expansion opportinity, the patent cliff approaching and #Neurodegeneration being completely underserved with huge unmet needs. It is inevitable in my mind...just like the recent Ferrari market which has exploded in the last year. What was an equitable and stable market can instantly change when scarcity value enters the equation. There is nothing like rexacerat, it is in a class by itself as the sole drug to ever show sustained disease modification in PD. Don't be surprised when you are asked to vote in the future. We have it and many will soon want it. I won't be surprised to soon see the shares better reflect this growing reality. Fine line between fear and greed and the pendulum tends to overswing...

$GANX Looks like Roche has been following our results closely and is sold on GCase biology. Today they paid $100M upfront + $1.1B milestones for a pre-clinical asset also targeting GCase biology. https://www.reuters.com/legal/litigation/alector-soars-roche-licenses-parkinsons-therapy-up-127-billion-2026-10-05/ Not even tested in humans yet and won’t be orally administered. It’s an engineered GCase enzyme so it may have a potency advantage in terms of restoring GCase. Personally think their money would’ve been much better spent on buying Gain which is already derisked and ph2-ready... but I think overall it’s a positive signal for our prospects and value.

$GANX About the time you want to puke, is about the time things reverse. We must be close based on my having to hold it down at least 3 times today. 😉 This will pass, always does, 25 years of experience doesn't make it any easier. This is where the professionals typically absorb retail holders supply as they capitulate. Large transference from weak hands to stronger, often followed by a steong move off the lows as emotions subside and fundamentals again come into focus. I suspect our pain may soon be pharma's gain. I expect this recent pressure in small biotech soon may fuel a pick up in acquisitions as valuations have reached to hard to pass up levels. $COYA

$GANX Zero transient liver enzyme elevations in the second part of the Phase 1b, and no other new safety signals. Not sure if this is new info, but it's more clear-cut than I recall — a really positive safety signal for an orally administered therapy, where the drug and its metabolites pass through the liver. Pleased to see this.

$GANX all high beta names are getting crushed... including the one that can prevent Parkinson’s Ive been buying gain particularly here over other names because of the confidende i have in our science, clinical data, and executive team. Market sucks right now but that's probably expected before midterm elections And rates are high not because the economy is bad. Its because massive demand for debt finances AI stuff is pulling demand away from government treasuries, etc. Its actully bullish but difficult for our little projects like GANX. There isnt infinite capital out there! Thats all We are so cheap - this could get sold for 10x overnight and it would be a steal of the decade for big pharma

$GANX I believe we can all agree, the only weakness of the company is the communication, at this point. We were under the impression the FULL dataset--(i.e--biomarker data), would be released at the Korea conf. & we still haven't been privy to. That doesn't concern me, if that wasn't expected. Just tell your shareholders that you're still validating the #'s or waiting for the last participant to finish the study. Simple. Say "we expect to release said data at the MJFF conference or another conference upcoming". Concrete information--i.e--release dates/information content with better communication, would go a long way to placate current shareholders/potential investors doubts. Otherwise, I'm not pessimistic at all. Patience pays here. Best to the longs....

$GANX Three M&A Fundamentals in Place The Patent Cliff: Over $300 billion in revenue (about 40% of total industry sales) is exposed to patent expirations over a five-year window, forcing companies to aggressively buy external pipelines. Spotty Internal Pipelines: Internal large pharma pipelines aren’t nearly strong enough to provide the growth drivers pharma needs to overcome upcoming patent cliffs. Over a Trillion Dollars of Buying Power: The pharma industry has approximately 1.2 trillion dollars in M&A buying power. Of course, only a fraction of this is going to get used but there is substantial capacity to buy the biotech market. This is not a theoretical topic. Our industry conversations aren’t telling us that M&A might pick up in the coming months. Those conversations tell us that M&A will pick up. And, with that, the biotech FOMO trade is likely to be right back.

$GANX Good info. I spoke with two execs i know from pharma last week after the overseas conference, they were there in person. they seemed impressed. from the m&a side. they were saying they are actively looking for bolt on deals in neuroscience. they didnt indicate whether gain is on the radar but they knew the drug and data well enough to make me assume they could be. the drug is a perfect acquisition target as a phase 2 ready asset. they did agree the ph1 is small but the antidotal evidence is strong regaining functions along with updrs scores is about the most encouraging data they have seen in pd. they liked the biomarkers and said that is something that pharma is interested in. they confirmed pharma is gun shy because many failures in the space but they think a breakthrough is coming. they knew i own the shares and said someone will probably buy them but it may be less than you hope for. that has been my worry for some time.

$GANX At least 1 presentation at Neuroscience 2026 by Joanne Taylor on preclinical backup compound.

$GANX I think if you can affect the upstream issues you can help with a large number of neurological diseases. Parkinsons and Alzheimers may have more in common than previously thought.

$GANX Not sure how this Novartis deal got by me. Looks like 575m upfront for a phase 1 asset.

$GANX Arrowhead puts out groundbreaking news and does a live Q and A with Management and doctors the same day. Our results were dumped into a poster at a conference and then silence. If GANX doesn't do something similar when they have the full 1 year data it won't be pretty. The market is deep red and this is a macro situation but the company needs to figure out how to communicate what we have!

$GANX I feel like we are all missing something, or not being told something by mgmt. How can a drug with such great data, this far advanced, in such a huge potential mkt perform so badly!!! And how can such an experienced mgmt team be so bad at communicating the message conference after conference and generate no interest??! The the the frick is going on?!?!?’

$GANX looks like they added smell tests for the preclinical mice tests after getting our human results.

$GANX $COYA you can tell how genuinely off the retail radar these companies are simply by the complete lack of trolls and bears on their respective pages…

$GANX I live in ireland and we once won the eurovision song contest with a song called What's another year. At this point i will take another year if the outcome is worth it. One things for sure. You won't be buying my shares if you add down here.

$GANX I had dinner with a BP exec last week and familiar with GT-02287. The gist is that he gave no credit to the efficacy and their models don’t account for that because it’s PH1, small, and not designed to measure that. I wonder how many other companies feel the same way. He did say that the fact it reached brain proved the mechanism works and warrants additional investment. Just not as bullish as us or the company.

$GANX We've been told that the UPDRS scores remain stable after one year. We haven't seen the biomarker data to back that up. However, the reverse would be more reason for doubt: there have been past attempts with drugs that showed effect in biomarkers, but did not translate into a change in the measurement of motor functioning or other real life indicators. Which is what the FDA is looking for first and foremost. It's hard to imagine how there could be a physical impact that does not show in the individual's biology. I'm inclined to believe that even a full 360 biomarker readout would not move the needle here. Only resolution of the Phase 2 funding, preferably involving a third party, will do that. All that being said, management should try to find ways to defend their capitalisation. Being stuck in penny stock land isn't helping anyone.
